Application
This kit can be used to measure CAT activity in animal serum, plasma, tissue, whole blood, red blood cells, culture cells, nutrient solution and other samples.
Detection principle
The reaction that catalase (CAT) decomposes H2O2 can be quickly stopped by ammonium molybdate. The residual H2O2 reacts with ammonium molybdate to generate a yellowish complex. CAT activity can be calculated by production of the yellowish complex at 405 nm.
Operation steps
1. Detection of CAT activity in serum(plasma)

Note: If there are no hemolysis and obvious blood liquid, the control tube can be replaced by double-distilled water for. Control tubes only need 1-2 for each experiment.
2. Detection of CAT activity in whole blood
Preparation of 1:99 hematolysis: Dilute 50 μL whole blood to 5 mL with double-distilled water. Mix fully and stand for 10 min.

Mix, then measure the OD values of each tube at 405 nm wavelength (using a cuvette of 0.5 cm diameter, set to zero with double-distilled water).
3. Detection of CAT activity in tissue homogenates
Sample preparation: Weigh the tissue accurately, add normal saline to tissue sample in a weight (g): volume (mL) ratio of 1:9,
prepare 10% tissue homogenate in ice water bath. Centrifuge the tissue homogenate for 10 min at 2500~3000 rpm. Collect the
supernatant and dilute it to the optimal concentration for detection.
